Sperm plays a crucial role beyond the simple transmission of paternal genetic information. The integrity of sperm DNA is essential to ensure the health of the offspring, and its evaluation should include specific DNA analyses. Traditional methods of semen analysis, which focus on sperm concentration, motility and morphology, are not reliable indicators of reproductive potential. In response, more advanced tests, notably those assessing DNA quality, have been developed. These tests show that sperm DNA damage is associated with various reproductive health problems, such as fertilization and spontaneous abortion.This review highlights sperm DNA analysis techniques, discussing their methodology, advantages, disadvantages and clinical implications. It also addresses the logistics involved in integrating these tests into the diagnosis of infertile men, as well as the challenges and opportunities they represent for assessing male fertility.
